Abstract

Epstein Barr virus (EBV) is an oncogenic herpesvirus aetiologically linked to several human malignancies of epithelial and lymphoid origins. An accumulating body of data indicates that EBV-infected cells release specific viral components in nano-vesicles called exosomes. Epstein-Barr virus encoded RNAs (EBER1 and EBER2) are two highly abundant, non-protein coding RNAs that are consistently expressed in all EBV latency programs. Conventional in situ hybridization (ISH) studies have indicated that these RNAs are typically found in the nucleus of EBV-infected cells. Recently, it was reported that EBERs are released out of the infected cells. However, the mechanism by which these highly abundant EBV products are transported from the nucleus to the cytoplasm and from there to outside of the cell remains unknown. In this thesis, we studied if EBER1 and EBER2 are excreted out from the infected cells via the exosomal pathway. Using differential ultracentrifugation, we isolated exosomes from EBV infected, non-infected and EBER1-transfected cell lines. The identity of purified exosomes was confirmed by electron microscopy and western blotting for exosomal markers. The presence of EBERs in cells, culture supernatant and purified exosomal fractions was determined using RT PCR. Both EBER1 and EBER2 were observed to be present not only in the culture supernatants but also in purified exosomes from all EBV-infected cell lines. The EBER binding protein La was also observed to be present in the purified exosomal fractions. To directly track the journey of these small RNAs, we developed a novel electron microscopy-based technique. Our results showed (for the first time) that at least a portion of EBERs are transported from the nucleus to the cytoplasm, where they appear to be loaded into multi-vesicular bodies for eventual excretion via exosomes. Furthermore, the EBER binding protein La was also localized to the same exosomal fractions, suggesting that EBERs might be released from the EBV-infected cell lines in the form of EBER-La complexes. Moreover, when the purified exosomes from both type I and type III EBV latently-infected cell lines were exposed to non-infected cells, they induced apoptosis in the recipient cells in a time and dose-dependent manner. The fluorescently-labeled exosomes isolated from EBV infected cell lines were taken up by non-infected cell lines, where they induced apoptosis via the extrinsic pathway of cell death. The blocking of caspase 3/7/8 pathways resulted in inhibition of exosome-mediated apoptosis. Molecular analysis of the exosomes from EBV-infected cell lines showed the presence of Fas ligand (FasL) in the exosomal fractions. Our data indicates that exosomes from EBV-infected cell lines induce apoptosis via the FasL-mediated extrinsic pathway and incubation of exosomes with anti-FasL antibody resulted in reduction of apoptosis in the recipient cells. Together, these data support the view that EBV can hijack the cellular exosomal pathway to excrete specific viral and cellular components to modulate the microenvironment.

فيروس "ابستين بار " (EBV) و من انواع الفيروسات المرتبطة بالعديد من الأورام الخبيثة التي تصيب الانسان في الخلايا الظهاريه واللمفاوية. وتشير المجموعة الهائلة من البيانات إلى ان الخلايا المصابة بفيروس .exosomes يعتبر EBER1,EBER 2 من الاحماض النووية الريبية المشفرة لفيروس EBV والمتوفرة بكثرة ويتم التعبير عنها دائما في المراحل الكامنة للفيروس. وقد أشارت الدراسات التقليدية باستخدام تقنية (ISH) ان هذه الاحماض النووية الريبية المشفرة (EBERs)عاده ما توجد في انوية الخلايا المصابة بفيروس ال .EBV في الاونه الاخيره، قد تبين بأنه يتم خروج ال EBERs من الخلايا المصابة. ومع ذلك ،فإن الآلية التي يتم بها نقل هذه الاحماض الوفيرة جدا من النواة إلى السيتوبلازم ومن هناك إلى خارج الخلية لا تزال غير معروفه. في هذه الاطروحه، درسنا إذا ما كانت ال EBER1 و EBER2 تفرز من الخلايا المصابة عبر مسارال .exosomes باستخدام تقنية ال ،ultracentrifugation تم عزل ال exosomes من الخلايا المصابة بال ،EBV والخلايا غير المصابة و خلايا ال .EBER1 transfected وتم التأكد من وجود ونقاء ال exosomes عن طريق المجهر الكتروني وتقنية ال western blotting تم تحديد وجود EBERS في الخلايا، وال culture supernatant وفي ال exosomes المعزولة عن طريق استخدام تقنية ال .RT PCR تم ايجاد كلا من ال EBER2 و EBER1 ليس فحسب في culture supernatant بل ايضا في ال exosomes المعزولة من كل الخلايا المصابة بفيروس ال .EBV ولوحظ أيضا ان البروتين الملزم لل La EBER يوجد ايضا في ال exosomes المعزولة. ولمتابعة الرحلة الشيقة لهذه الاحماض الريبية الصغيرة، طورنا تقنيه جديدة ومستحدثة باستخدام المجهر الاكتروني. وأظهرت نتائجنا للمرة الاولي ان نسبة من ال EBERS يتم نقلها من النواة إلى السيتوبلازم ، حيث وعلى ما يبدو انه يتم تحميلها في اجسام متعددة الجذور لإفرازها في نهاية المطاف عن طريق .exosomes وعلاوة علي ذلك، فان البروتين الملزم لل

، EBER La كان أيضا موجودا في نفس المحتوى لل exosomes مما يوحي بأنه قد يتم الإفراج عن EBER من الخلايا المصابة بال EBV على هيئة .EBER-La وعلاوة علي ذلك، فإن ال .exosomes ( المستخرجة من كل من النوع الأول والنوع الثالث للخلايا المصابة بفيروس ال EBV ) عند تعريضها للخلايا غير المصابة ، فانها تؤدي إلى حفز موت الخلايا المبرمج في الخلية المتلقية اعتمادا على الوقت والجرعة. وعلاوة علي ذلك، فإن ال exosomes المعالجة بالاشعه الفلورية والتي تم عزلها من الخلايا المصابة بالEBV ، تم اكتسابها من قبل الخلايا غير المصابة، حيث انها تسببت في موت الخلايا المبرمج عن طريق المسار الخارجي لموت الخلية. أدى وقف مسارات الكاسباس 8/7/3 الى تثبيط موت الخلايا المبرمج عن طريق ال exosomes وأظهرت التحاليل الجزيئية لل exosomes الى وجود رابط ال (FasL)FAS في محتوى ال .exosomes وأشارت بياناتنا الى ان ال exosomes تسبب الموت المبرمج للخلايا عن طريق المسار الخارجي لرابط ال FasL وان وضع ال exosomes مع مضاد ال FasL يؤدي الى الحد من الموت المبرمج للخلية المتلقية. مع اخذ كل هذه النتائج في الاعتبار، فإن بياناتنا تدعم الراي القائل بان ال EBV يمكنه خطف المسار الخلوي لل exosomes لإفراز مكونات محددة فيروسية وخلوية لتحوير البيئة من حوله.
