Abstract

Acrylamide is a toxic compound that is formed in heated carbohydrate-rich food. Scientists have analyzed acrylamide levels before, during, and after processing in order to study the mitigation strategies and, due to the lack of knowledge, this study aimed to: 1) screen 120 newly isolated LAB for their acrylamide removal; 2) optimize the conditions for 6 selected strains of pH, temperature, time and salt, using the Box-Behnken design (BBD), and analyze their acrylamide removal levels; 3) investigate acrylamide removal abilities of the selected LAB isolates under in-vitro digestion conditions, using the INFO-GEST 2.0 model; and 4) examine the mechanism of acrylamide removal, using scanning electron microscopy coupled with energy-dispersive X-ray spectroscopy (SEM-EDS), zeta potential, transmission electron microscopy (TEM) measurement, and Fourier transform infrared spectroscopy (FTIR). Out of the 120 strains, the highest six in acrylamide removal were Streptococcus lutetiensis, Lactiplantibacillus plantarum, Bifidobacterium breve, Lactiplantibacillus plantarum, Enterococcus durans and Enterococcus faecalis, with reduction percentages of 39%, 26%, 35%, 36%, 33% and 30%, respectively. To enhance the binding ability of the strains, they were tested under controlled conditions of pH (4.5, 5.5 and 6.5), temperature (32°C, 37°C, and 42°C), time (14, 18 and 22 hours), and NaCl (0%, 1.5% and 3% w/v), using the Box-Behnken design (BBD). Strains removed more acrylamide in the range of 35-46% for S. lutetiensis, 45-55% for L. plantarum, 47.7-65.0% for B.breve and 5.8 – 38.0% for Lb. plantarum, 44% for E. durans and E. faecalis for 53%. After testing the bacterial binding ability, strains were exposed to a simulated gastrointestinal tract environment, with both S. lutetiensis and L. plantarum removing more than 30% of acrylamide at the gastric stage and around 40% at the intestinal stage. Under the in-vitro gastrointestinal conditions, B. breve removed up to 41.2% of acrylamide, while Lb. plantarum reduced around 53.5% of acrylamide. E. durans and E.faecalis maintained their binding ability under the simulated gastrointestinal environment, with removal percentages of 44% and 53%, respectively. To understand the mechanism of removal, LAB cells were characterized via scanning electron microscopy, coupled with energy-dispersive X-ray spectroscopy (SEM-EDS) and transmission electron microscopy (TEM) techniques. Cell charges were characterized by zeta potential and functional groups analyzed by Fourier transfer infrared spectroscopy (FTIR). The results indicated that FTIR and EDS diagrams and data presented the association of the functional groups C=O, C-O, and N-H in the acrylamide adsorption process. TEM results indicated that the cell walls of the bacteria increased when cultured in media supplemented with acrylamide. This study revealed that bacterial species and strain variations affect acrylamide removal capabilities. This study indicated that lactic acid bacteria and bifidobacteria might be an ambitious solution that can be used successfully in removing toxins in different applications. In this study, we used the bacteria to remove the acrylamide successfully in-vitro in liquid MRS broth and in a simulated gastrointestinal condition with a new method and newly isolated strains of LAB. Further studies are needed to examine LAB behavior in vivo conditions in the human body and regular diets.

الأكريلاميد هو مركب سام يتكون في الأطعمة المطبوخة الغنية بالكربوهيدرات، وقد قام العلماء بتحليل مستويات مادة الأكريلاميد قبل وأثناء وبعد الطهي من أجل دراسة طرق تقليل تشكل الأكريلاميد في الأطعمة، وبسبب نقص المعرفة، هدفت هذه الدراسة إلى:1) فحص قدرة 120 بكتيريا حمض اللاكتيك معزولة حديثاً لإزالة مادة الأكريلاميد؛ 2) اختيار 6 بكتيريا وتحسين الظروف المحيطة بها لرفع مستوى أدائها من الأس الهيدروجيني ودرجة الحرارة والوقت والملح باستخدام تصميم Box-Behnken BBD، 3) التحقق من قدرة إزالة مادة الأكريلاميد تحت ظروف الهضم في المختبر باستخدام نموذج INFO-GEST 2.0، 4) فحص آلية إزالة مادة الأكريلاميد باستخدام الفحص المجهري الإلكتروني المقترن بمطياف الأشعة السينية المشتت للطاقة (SEM-EDS)، وإمكانات زيتاً، وقياس المجهر الإلكتروني للإرسال (TEM)، والتحليل الطيفي للأشعة تحت الحمراء (FTIR). من بين 120 سلالة، كانت أعلى ست سلالات في إزالة مادة الأكريلاميد هي Streptococcus lutetiensis، Lactiplantibacillus plantarum، Bifidobacterium breve، Lactiplantibacillus plantarum، Enterococcus durans، Enterococcus faecalis بنسب تخفيض 39%، 26%، 35%، 36%، 33%، و30% على التوالي. لتعزيز قدرة السلالات البكتيرية، تم اختبارها تحت الظروف التالية: الأس الهيدروجيني (4.5، 5.5، 6.5)، ودرجة الحرارة (32 درجة مؤوية، 37 درجة مؤوية، 42 درجة مؤوية)، والوقت (14، 18 و22 ساعة)، والملح NaCl (0%، 1.5%، 3% وزن/حجم) باستخدام تصميم Box-Behnken (BBD). السلالات البكتيرية أزالت كميات أكريلاميد متفاوتة في النطاقات التالية: S. lutetiensis من 35%-46%، L. plantarum من 45%-55%، B.breve من 47.7%-65.0%، Lb. plantarum من 5.8%-38.0%، E. durans بنسبة 44% ، وأخيراً E. faecalis بنسبة 53%.

بعد اختبار قدرة الارتباط البكتيري، تم فحص السلالات البكتيرية في بيئة مشابهة للجهاز الهضمي في جسم الإنسان، حيث تمكنت كل من L. plantarum و S. lutetiensis من إزالة أكثر من 30% من مادة الأكريلاميد في مرحلة الهضم في المعدة وحوالي 40% في المرحلة المعوية. في ظل ظروف الجهاز الهضمي في المختبر، أزالت B.breve ما يصل إلى 41.2% من مادة الأكريلاميد، بينما أزال Lb. plantarum حولي 53.5% من مادة الأكريلاميد. حافظت E. durans و E. faecalis على قدرتها على ربط مادة الأكريلاميد تحت هذه الظروف بنسب إزالة 44% و53%.

لفهم آلية عمل البكتيريا، تم تمييز الخلايا عن طريق المسح المجهري الإلكتروني، إلى جانب تقنيات التحليل الطيفي للأشعة السينية المشتتة للطاقة (SEM-EDS) وتقنيات النقل المجهري الإلكتروني (TEM) إلى جانب فحص شحنات الخلايا بإمكانية زيتا وفحص المجموعات الوظيفية بواسطة مطياف فورييه بنقل الأشعة تحت الحمراء (FTIR). أشارت النتائج إلى أن مخططات وبيانات FTIR وEDS تفيد بارتباط المجموعات الوظيفية C = O و C-O و N-H في عملية ربط الأكريلاميد بجدار الخلايا، كما أشارت نتائج TEM إلى أن أن جدران الخلايا البكتيرية تزداد عند زراعتها في وسط يحتوي على مادة الأكريلاميد، إضافة إلى ذلك فقد أشارت هذه الدراسة إلى أن الأنواع البكتيرية واختلافات السلالات تتفاوت في قدرتها على إزالة مادة الأكريلاميد.

أشارت هذه الدراسة إلى أن بكتيريا حمض اللاكتيك والبكتيريا المشقوقة قد تكون حلاً طموحاً يمكن استخدامه بنجاح في إزالة السموم في مختلف التطبيقات وفي هذه الدراسة، استخدمنا البكتيريا لإزالة الأكريلاميد بنجاح في المختبر في وسط الزرع MRS السائل وتعريض سلالات بكتيرية معزولة حديثًا لبيئة مشابهة للجهاز الهضمي في جسم الإنسان وأيضًا تمكنت من إزالة الملوث الكيميائي الأكريلاميد بنجاح. هناك حاجة إلى مزيد من الدراسات لفحص سلوك بكتيريا حمض اللاكتيك في ظروف جسم الإنسان بوجود الوجبات الغذائية الاعتيادية التي يتناولها الإنسان للتحقق من إمكانية البكتيريا من إزالة السموم في مختلف الظروف.
