Abstract
Plastics, including microplastics, pose significant environmental threats due to their widespread presence and harmful ecological effects. This dissertation present three distinct, yet interconnected studies. First, a study on microplastic contamination in urban recreational parks was conducted in Al Ain, United Arab Emirates. Results showed that low-density microplastics were present in 87% of soil samples, with spectroscopic analysis identifying synthetic polymers such as polyethylene and polypropylene. Additionally, these microplastics were found to negatively affect soil pH and moisture levels, emphasizing the need for enhanced monitoring and management strategies. Second, the effects of ground microplastics on Photobacterium leiognathi strains, a marine luminous bacterium, were examined. Findings revealed that microplastic exposure altered bioluminescence, cell viability, and influenced biofilm formation. Moreover, strain-specific responses indicated that ground microplastics disrupted quorum sensing, suggesting that these plastic particles act as environmental stressors on marine microorganisms. Lastly, to address polyethylene terephthalate persistence, synthetic microbial consortia or microbial communities were developed. These consortia demonstrated potentially for enhance polyethylene terephthalate degradation via synergistic microbial interactions, biofilm formation, and enzymatic activity. The study also conducted whole genome sequencing of two bacterial strains, Bacillus safensis F18 and Escherichia coli 9SH, that have potential application in plastic biodegradation. The B. safensis F18 possessed known plastic-degrading enzyme called hydrolase. Therefore, these findings offer promising strategies for mitigating plastic pollution through microbial degradation. Overall, this dissertation contributes to the understanding of microplastic pollution impacts and offer promising bioremediation approaches, with potential applications in industrial-scale waste management.

من تأثير البلاستيك الدقيق إلى حلول التحلل البيولوجي: التحقيق في تلوث البلاستيك الدقيق، والاستجابات الميكروبية، وفعالية اتحادات الميكروبات الاصطناعية في تحلل البلاستيك
تُشكل المواد البلاستيكية، بما فيها الجسيمات البلاستيكية الدقيقة، تهديدات بيئية جسيمة نظرًا لانتشارها الواسع وتأثيراتها البيئية الضارة. تُقدم هذه الرسالة ثلاث دراسات مُتميزة، وإن كانت مترابطة. أولًا، أُجريت دراسة حول تلوث المواد البلاستيكية الدقيقة في الحدائق الترفيهية الحضرية في مدينة العين، الإمارات العربية المتحدة. أظهرت النتائج وجود جسيمات بلاستيكية دقيقة منخفضة الكثافة في 87% من عينات التربة، حيث كشف تحليل فوريه تحويل للأشعة تحت الحمراء عن وجود بوليمرات صناعية مثل البولي إيثيلين والبولي بروبيلين. بالإضافة إلى ذلك، وُجد أن هذه الجسيمات البلاستيكية الدقيقة تؤثر سلبًا على درجة حموضة التربة ومستويات رطوبتها، مما يؤكد الحاجة إلى استراتيجيات مُحسّنة للرصد والإدارة. ثانيًا، تم فحص آثار الجسيمات البلاستيكية الدقيقة على بكتيريا Photobacterium leiognathi strains. كشفت النتائج أن التعرض للجسيمات البلاستيكية الدقيقة يُغيّر من التألؤ الحيوي، وقابلية الخلايا للحياة، ويؤثر على تكوين الأغشية الحيوية. علاوة على ذلك، أشارت الاستجابات الخاصة بكل سلالة إلى أن الجسيمات البلاستيكية الدقيقة الأرضية تُعطل استشعار النصاب، مما يُشير إلى أن هذه الجسيمات البلاستيكية تُشكل ضغوطًا بيئية على الكائنات الحية الدقيقة البحرية. أخيرًا، لمعالجة مشكلة ثبات البولي إيثيلين تيريفثالات، طُوّرت اتحادات ميكروبية اصطناعية أو مجتمعات ميكروبية. وقد أثبتت هذه الاتحادات قدرتها على تعزيز تحلل البولي إيثيلين تيريفثالات من خلال التفاعلات الميكروبية التآزرية، وتكوين الأغشية الحيوية، والنشاط الإنزيمي. كما أجرت الدراسة تسلسلًا كاملًا للجينوم لسلالتين بكتيريتين، هما Bacillus safensis F18 و Escherichia coli 9SH، واللتان يُمكن تطبيقهما في التحلل البيولوجي للبلاستيك. تمتلك B. safensis F18 إنزيمًا معروفًا لتحلل البلاستيك يُسمى هيدرولاز. لذلك، تُقدم هذه النتائج استراتيجيات واعدة للتخفيف من تلوث البلاستيك من خلال التحلل الميكروبي. وبشكل عام، تُسهم هذه الأطروحة في فهم آثار تلوث البلاستيك الدقيق، وتُقدم مناهج واعدة للمعالجة البيولوجية، مع تطبيقات محتملة في إدارة النفايات الصناعية.
